AMPLY Discovery is a UK biotech company focused on discovering novel RNA medicines for oncology. The company targets two conventionally undruggable cancer types: triple-negative breast cancer and high-grade serous ovarian cancer, using a platform called TALON that integrates six specialized machine learning engines for target discovery, validation, and therapeutic design.

The TALON platform combines computational prediction with wet-lab validation across two translational hubs—one at Queen's University Belfast and one at City St George's, University of London. The company has three primary programmes in development: AMPLY-LNC1 (targeting long non-coding RNA in triple-negative breast cancer), AMPLY-LNC2 (targeting long non-coding RNA in high-grade serous ovarian cancer), and AMPLY-TF1 (targeting a master transcription factor in high-grade serous ovarian cancer). All three programmes are currently at the in vitro proof-of-concept stage.

Notable finds

  • EXPLORER — Atlas-scale lncRNA target discovery with Cancer-specific dependency scoring
  • HALLUX — Multi-omic ML triage with Twelve disqualifying gates for tractability and druggability assessment
  • MtG — Gapmer ASO Therapeutic Design for nuclear lncRNA knockdown
  • siREN — Generative siRNA design for cytoplasmic RNA targets
  • THEBOX — Browser-native lncRNA custom primer designer for direct-lysate RT-qPCR
  • FOLIO — Tolerability and toxicity prediction engine trained on 1,134 measured HC50s
  • Wet-lab validation in two translational hubs at Queen's University Belfast and City St George's
  • Three independently gated oncology programmes targeting TNBC and ovarian cancer

Questions from the trail

What makes triple-negative breast cancer and ovarian cancer conventionally undruggable?

Triple-negative breast cancer and high-grade serous ovarian cancer have two major target classes that traditional drug modalities cannot reach: long non-coding RNAs (which produce no protein for antibodies or small molecules to bind) and master transcription factors (which have no druggable pocket and have paralogues too similar for protein-level selectivity). RNA medicines overcome this by reading sequence rather than shape, allowing a single chemistry to reach both target types.

What is TALON and how does it work?

TALON is AMPLY's integrated machine learning discovery platform consisting of six specialized engines. Each engine solves one part of the dark-genome discovery problem: EXPLORER for target discovery, HALLUX for target validation, MtG for antisense design, siREN for siRNA design, THEBOX for wet-lab primer design, and FOLIO for toxicity prediction. Together they form an end-to-end stack for discovering targets the field has missed.

How many programmes does AMPLY have in development and what is their status?

AMPLY has three oncology programmes in development, each at the in vitro proof-of-concept stage. AMPLY-LNC1 targets a long non-coding RNA in triple-negative breast cancer; AMPLY-LNC2 targets a long non-coding RNA in high-grade serous ovarian cancer; and AMPLY-TF1 targets a master transcription factor in high-grade serous ovarian cancer. Each programme gates independently so one programme failing does not take the others with it.

Where are AMPLY's wet-lab facilities located?

AMPLY operates two translational hubs. The Belfast hub at Queen's University Belfast focuses on discovery and validation work through the Future Medicines Institute and the Patrick G. Johnston Centre. The London hub at City St George's, University of London focuses on toxicity, imaging, and peptide chemistry work through the Institute for Infection & Immunity.
